چکیده
In this research, immobilization of A. fumigatus ?-amylase on chitin was studied with the main purpose to improve characteristics enzyme. A series experiments were carried out study stability improvement, thermodynamic parameters, include ki, ?Gi, and t½, reusability immobilized The experimental results indicate that significant thermal achieved, as indicates by ability enzyme retain its relative activity above 39% after 80 min incubation at 60oC. Thermodynamic is more rigid, stable, less flexible in water, resulting increased up 1.5 times compared native Furthermore, able over 46% initial six consecutive applications for starch hydrolysis, confirming potential production enzymes an industrial scale. Chitin biosynthesis in protoplasts and subcellular fractions of Aspergillus fumigatus.
The biosynthesis of chitin has been obtained in broken mycelia and protoplasts of the fungus Aspergillus fumigatus. The specific activity of chitin synthase (EC 2.4.1.16) in a membrane preparation from protoplasts derived from the hyphal tips of A. fumigatus was 26.8-fold greater than that of the chitin synthase in broken mycelia, indicating that the active chitin synthase is located primarily ...
متن کاملOptimization and immobilization kinetics of Aspergillus nidulans cellulase onto modified chitin by response surface approach
The present study deals with the immobilization of Aspergillus nidulans cellulase onto modified chitin (MC). The effect of contact time, cellulase concentration, MC dosage, temperature and agitation rate for maximum immobilization percentage and immobilization capacity is investigated on the basis of five-level-five-factor central composite design (CCD) obtained by response surface methodology ...
متن کاملBiofilm formation by Aspergillus fumigatus.
Aspergillus fumigatus is a well adapted, opportunistic fungus that causes a severe and commonly fatal disease, invasive pulmonary aspergillosis (IPA), in highly immunocompromised patients, aspergilloma in patients with lung cavities and allergic bronchopulmonary aspergillosis (ABPA) in hypersensitive individuals. Recent studies have suggested that biofilm formation by A. fumigatus may be one of...
متن کاملCaspofungin Treatment of Aspergillus fumigatus Results in ChsG-Dependent Upregulation of Chitin Synthesis and the Formation of Chitin-Rich Microcolonies
Treatment of Aspergillus fumigatus with echinocandins such as caspofungin inhibits the synthesis of cell wall β-1,3-glucan, which triggers a compensatory stimulation of chitin synthesis. Activation of chitin synthesis can occur in response to sub-MICs of caspofungin and to CaCl2 and calcofluor white (CFW), agonists of the protein kinase C (PKC), and Ca(2+)-calcineurin signaling pathways. A. fum...
متن کاملCo-recognition of β-glucan and chitin and programming of adaptive immunity to Aspergillus fumigatus
The prevalence of fungal infections has increased concurrently with increases in immune suppressive therapies and susceptible individuals. Opportunistic fungal pathogens such as Aspergillus fumigatus may exhibit invasive growth and dissemination resulting in a high mortality rate. Herein, we discuss how immune sensing of germination directs innate immune responses and programs adaptive response...
